Abstract

식물생장촉진세균은 식물의 생장과 수확량을 촉진하고, 식물병에 대한 유도저항성을 유도하는 것으로 보고되었다. 본 논문에서 연구의 목적은 가로수와 고추의 엽면에 엽권정착 식물생장촉진세균을 처리하여, 식물생장촉진세균의 적용 범위를 확장하였다. 수목의 엽권에서 내생포자 형성 세균 1,056개 균주를 분리하여, protease, chitinase, lipase를 포함한 효소활성과 진균병인 C. graminicolaB. cinerea에 대한 길항작용을 측정하였다. 1차 선발된 bacilli 14개 균주를 고추의 잎에 살포하여 엽권정착능을 시험하였다. 5B6, 8D4, 8G12 단독처리와 그 혼합처리군을 고추 엽면에 살포하여 생장촉진, 수확량증진, 병방제 효과를 고추 포장에서 관찰하였다. 대량배양을 통하여 선발된 균주를 대한민국 대전광역시 유성구 일대의 가로수에 살포하였을 때, 대조군과 비교하여 엽록소함량과 잎 두께가 증가하였다. 선발된 3개 균주를 수목에 엽면살포했을 때, 벚나무 진균성갈색무늬구멍병을 저해하였고 은행나무의 낙엽생성을 촉진하였다. 종합적으로 본 연구는 엽권정착세균의 엽면살포를 통하여 가로수와 고추의 생장을 촉진시키고, 식물병을 방제하는 엽권정착세균의 적용 가능성을 제시한다.

Trans Abstract

Out of plant-associated bacteria, certain plant growth-promoting bacteria (PGPB) have been reported to increase plant growth and productivity and to elicit induced resistance against plant pathogens. In this study, our objective was to broaden the range of applications of leaf-colonizing PGPB for foliar parts of road tress and pepper. Total 1,056 isolates of endospore-forming bacteria from tree phylloplanes were collected and evaluated for the enzymatic activities including protease, lipase, and chitinase and antifungal capacities against two fungal pathogens, Colletotrichum graminicola and Botrytis cinerea. Fourteen isolates classified as members of the bacilli group displayed the capacity to colonize pepper leaves after spraying inoculation. Three strains, 5B6, 8D4, and 8G12, and the mixtures were employed to evaluate growth promotion, yield increase and defence responses under field condition. Additionally, foliar application of bacterial preparation was applied to the road tress in Yuseong, Daejeon, South Korea, resulted in increase of chlorophyll contents and leaf thickness, compared with non-treated control. The foliar application of microbial preparation reduced brown shot-hole disease of Prunus serrulata L. and advanced leaf abscission in Ginkgo biloba L. Collectively, our results suggest that leaf-colonizing bacteria provide potential microbial agents to increase the performance of woody plants such as tree and pepper through spray application.

서론

식물은 지구 상에서 성공적으로 정착한 생물종이고, 식물의 지상부는 지표면의 2배 이상을 차지할 만큼 넓은 표면적을 갖고 있다(Lindow와 Brandl, 2003; Lindow와 Leveau, 2002). 식물은 잎, 뿌리, 꽃, 종자, 열매와 같이 미생물이 정착할 수 있는 표면을 제공하고, 많은 경우 식물이 표면정착세균과 상호작용을 하는 것으로 보고되었다(Lugtenberg 등, 2002). 식물은 구조상 크게 지상부와 지하부로 구분할 수 있는데, 표면정착세균의 정착 장소에 따라 각각 지상부의 엽권과 지하부의 근권으로 크게 나눌 수 있다(Leveu, 2015). 최근 연구동향을 보면 근권에 정착하는 미생물에 대한 연구는 활발하게 이루어졌으나 엽권에 정착하는 미생물에 대한 연구는 상대적으로 미진한 상황이다(Cirvilleri 등, 2008; Delmotte 등, 2009; Knief 등, 2012). 엽권정착세균(phyllobacteria)은 표면정착세균 중에서 엽권에 정착하여 살아가는 세균을 총칭하며, 생장촉진과 같이 식물에 긍정적인 영향을 주거나 또는 지상부에 병을 일으키는 것과 같은 부정적인 영향도 주며 엽권에 정착하여 살아간다(Glick, 2012; Lindow와 Brandl, 2003; Vorholt, 2012). 식물의 표면에 정착하여 식물에 긍정적인 영향을 주는 세균 군집 중에서 식물의 생장을 촉진시키는 세균을 총칭하여 식물생장촉진세균(plant growth-promoting bacteria, PGPB)으로 명명하였다(Glick, 2012; Glick과 Bashan, 1997; Vorholt, 2012). PGPB가 직•간 접적으로 유해한 미생물을 저해하고 식물의 유도전신저항 성(induced systemic resistance, ISR)을 일으키는 효과가 보고되면서, PGPB를 식물에 처리하여 화학농약의 단점을 극복하는 친환경적인 병 방제방법으로 인식하게 되었다(Bashan과 Holguin, 1998; Compant 등, 2005; Whipps, 2001). 방법론적으로는 PGPB로 알려진 Bacillus속이나 Pseudomonas속을 근권에서 분리하여 식물의 뿌리에 처리하는 방법이 주로 시도되었다(Bhattacharyya와 Jha, 2012; Kishore와 Pande, 2007; Kloepper 등, 1980). 최근에는 PGPB의 적용방법을 확대하기 위해서 PGPB를 작물이나 목본식물의 엽면에 살포하는 방식도 시도되었다(Baker 등, 1983; Chung 등, 2008; Jiang 등, 2006; Korsten 등, 1997; Lee 등, 2008; Obradovic 등, 2004; Pusey, 1989; Raupach와 Kloepper, 1998; Silva 등, 2004). 토양에서 분리한 P. fluorescensP. putida를 수목에 엽면살포한 결과 사과의 검은별무늬병을 방제하였으며(Ganeshan과 Manoj Kumar, 2005; Kinkel, 1997; Kucheryava 등, 1999; Planchamp 등, 2015), 사과나무 근권에서 분리한 PGPB를 사과나무에 엽면살포한 결과 수목의 생장이 촉진되고 수확량이 증대하는 사례도 보고되었다(Ryu 등, 2011). 또한, 토양에서 분리한 Bacillus subtilis Osu-142 균주를 살구수목에 살포하였을 때 살구나무의 생장과 수확량이 증진되는 효과와 진균병인 Wilsonomyces carpophilus에 대한 방제 효과도 보고되었다(Altindag 등, 2006; Çkmakıı 등, 2001; Eşitken 등, 2002; Esitken 등, 2006). 과실수, 조경수로 널리 이용되는 벚나무속(Prunus spp.)의 경우, PGPB인 Pseudomonas BA-8와 Bacillus OSU-142를 엽면에 살포하여 생장촉진과 수확량 증대효과를 확인했지만(Esitken 등, 2006), 벚나무속의 주된 병인 벚나무 진균성갈색무늬구멍병(Blumeriella jaapii)에 대해서는 항진균제인 sterol demethylation inhibitor를 이용한 화학적 방제가 주가 되었고, PGPB를 이용한 생물적 방제는 거의 시도되지 않았다(Eşitken 등, 2002; Ma 등, 2006).

본 연구에서는 수목의 지상부 병을 방제하기 위한 방법으로 엽권에 정착하는 PGPB 처리에 의한 수목의 생장촉진 효과와 병저항성 반응을 연구하였다. 엽권정착능이 뛰어난 bacilli 계열 균주가 내생포자를 형성한다는 특성을 이용하여 생물적 방제제로서 장기보관과 운반이 용이하다는 장점을 확보하였다. 분리한 엽권정착세균의 가로수에서의 활성을 확인하기 앞서, 가로수와 유사하게 목질부를 형성하는 특징을 가지고 있는 고추(Capsicum annuum L.)를 목본식물의 모델로 선정하여(Zhigila 등, 2014), 선발된 PGPB의 효과를 확인하였다. 또한 PGPB에 의한 은행나무 낙엽생성에 대한 연구도 진행하여 종합적인 가로수 관리의 가능성을 탐색하였다.

재료 및 방법

엽권정착 내성포자세균의 분리

수목의 엽권에 정착하는 내생포자생성 세균만을 분리하기 위해서 대한민국 대전광역시 유성구 일대의 벚나무(Prunus serrulata L.) (위도: 36.3616656°, 경도: 127.3565759°), 은행나무(Ginkgo biloba L.) (위도: 36.3766593°, 경도: 127.3613144°), 이팝나무(Chionanthus retusus) (위도: 36.3627869°, 경도: 127.3448559°) 잎을 무작위로 채취했다. 코르크보러(cork borer, 직경 10 mm)를 이용하여 무작위로 수거한 나뭇잎에서 지름 10 mm의 잎조각을 얻었다. 획득한 잎조각을 멸균된 1.0 ml phosphate buffered saline (PBS)가 담긴 1.5 ml 튜브에 넣은 후, 플라스틱 막자로 잎조각을 갈아주었다. 내생포자를 형성하는 세균만을 분리하기 위하여 80°C water bath에 30분간 중탕처리를 하였다. 중탕된 잎조각이 포함된 PBS 완충액을 1/10 tryptic soy agar (TSA) 배지에 도말한 후 30°C 배양기에서 12시간 배양을 한 후에, 배양되어 나온 단일 세균 집락을 형태학적 형질 및 표현형에 따라 분리하였다. 분리된 1,056개 균주는 96-well plates로 –80°C 극저온냉동고에 보관하였다.

엽권정착 내성포자생성 균주의 생물학적 활성 분석 및 균주 동정

분리된 1,056개 균주는 생물학적 활성을 측정하기 위해서 protease, chitinase, lipase 활성을 측정하였다. Protease 활성은 배지(1% skim milk, 0.02% sodium azide, 2.0% agar)에서 측정하였고(Chantawannakul 등, 2002), chitinase 활성은 배지(5% v/v chitin, 0.7 g K2HPO4, 0.3 g KH2PO, 0.5 g MgSO4ㆍ5H2O, 0.01 g FeSO4ㆍ7H2O, 0.001 g ZnSO4, 0.001 g MnCl2, 1.5% agar)에서 측정하였고(Hsu와 Lockwood, 1975), lipase 활성은 배지(1.0 g yeast extract, 5.0 ml corn oil, 2.0 g NaCl, 0.4 g MgSO4ㆍ7H2O, 0.7 g MgCl2ㆍ6H2O, 0.5 g CaCl2ㆍ2H2O, 0.3 g KH2PO4, 0.3 g K2HPO4, 0.5 g (NH4)2SO4, 1.0 ml vitamin solution [DSM catalogue No. 141], 1.0 ml trace element solution [DSM catalogue No. 141], 1.5% agar)에서 측정하였다(Lee 등, 1999). 각 효소의 활성을 대량 스크리닝으로 확인하기 위해 96 well plate 상태로 보관된 엽권정착 균주를 96-well microplates stamp를 이용해 250×250×11 mm2 크기의 사각 플레이트(square dish; SPL, Suwon, Korea)에 찍어서 접종하였다. 이후 30°C 배양기에서 12시간 배양을 한 후에 각 배지의 기질이 분해되면서 생기는 환의 생성 유무를 측정하여 생물학적 활성을 결정하였다.

효소활성 측정 방식과 동일한 방법으로 엽권에서 분리한 균주를 potato dextrose agar (24 g/l potato dextrose broth [DifcoTM; BD Diagnostic Systems, Sparks, MD, USA], 2.0% agar)에 접종한 후에 30°C에서 배양한 식물병원균 Botrytis cinerea (KACC accession No. 40573; Han 등, 2012), Collectotricum graminicola (Behr 등, 2010)의 배양액을 250×250×11 mm2 크기의 사각 플레이트에 각각 1 ml씩 도말하였다. 이후 30°C 배양기에서 1–2일 동안 배양한 후에, 엽권정착 균주에 의해 발생한 환의 생성 유무를 통해 길항작용의 유무를 판단하였다.

분리된 PGPB 균주의 엽권정착능 측정

분리된 14개의 엽권정착 균주를 항생제인 리팜피신(Rifampicin ≥97% [HPLC] powder; Sigma-Aldrich, St. Louis, MO, USA)의 농도 50 μ/ml가 포함된 TSA 배지에서 키워 리팜피신 저항성을 획득하도록 유도하였다. TSB 배지에 30°C에서 8시간 동안 배양한 후, 4엽까지 성장한 고추의 엽권에 OD600=1로 세균 집락의 농도를 맞춰준 후, 분무기를 이용하여 고추 엽권에 배양액을 살포하였다. 7일째와 10일째 균주를 살포한 고추의 잎을 3개씩 다시 수거하여 코르크보러를 이용하여 10 mm 직경의 잎조각을 각 균주 처리군별로 3개씩 3반복을 얻는다. 얻은 잎조각을 0.1 M MgCl2가 담긴 1.5 ml tube에 넣고 호모지나이저(homogenizer)를 이용하여 갈아준 후, 리팜피신이 포함된 TSA 배지에 농도에 따라 순차적으로 도말(serial dilution)한 후 세균 집락의 개수를 측정하였다.

선발된 PGPB를 이용한 고추의 생물학적 활성 측정을 위한 포장 실험

고추 포장실험은 충청북도 청원군 소재(위도: 36.353227°, 경도: 127.303475°) 고추밭에서 행해졌다. 13개의 분리된 균주 중 복수 활성을 보이는 3개 균주를 TSA 배지에 배양한 후에, 107 cfu/ml의 농도로 고추 엽면에 처리군(5B6, 8D4, 8G12, 혼합처리), 0.1 mM BTH, 대조군별로 완전임의화설계(30개체씩 4반복)를 하였으며, 10일 간격으로 2회, 1개체당 25 ml씩 살포하였다. 이후 20일 간격으로 고추의 길이를 60일까지 측정하였고, 동시에 Xanthomonas axonopodis pv. vesicatoria (Yi 등, 2012)에 대한 병저항성을 측정하기 위해서 OD600=1 농도의 균주를 PGPB 처리 20일 후 고추의 한 개체, 처리일 기준으로 각 3개의 잎에 주사기로 접종하였다. 이후 20일 간격으로 80일까지 병발생정도(0, 병징 없음; 1, 물기를 머금은 듯한 병징; 2, 잎표면에 작은 점무늬 병반 발생; 3, 많은 점무늬 병반의 발생과 시듦병 증세의 시작; 4, 점무늬 병반이 잎표면을 뒤덮고, 접종한 잎의 시듦이 본격화됨; 5, 접종한 잎의 완전 괴사)를 측정하였다(Sahin과 Miller, 1998). 고추의 수확량 측정을 위해서 PGPB를 살포한 날짜를 기준으로 64일(1차)과 80일(2차)에 고추의 열매를 수확하여 처리군별로 수확한 고추 개수를 무게로 나눈 평균값과 누적평균값을 측정하였다.

분리된 PGPB 균주의 가로수 적용 실험 및 생물학적 활성 검정

분리된 3균주의 벚나무, 은행나무, 이팝나무에서의 활성을 측정하기 위해 3균주를 기반으로 한 미생물제제를 제작하였다. 3균주(5B6, 8D4, 8G12)의 내생포자 분말을 공업적으로 대량배양한 후, 정착능을 높이기 위해서 전착제인 실루엣(20 l; Agrotech, Seoul, Korea)을 균액 20 l당 6.7 ml 첨가하여 준비하였다(Ryu 등, 2011). 대전광역시 일대의 벚나무, 은행나무, 이팝나무에 대량 배양한 3균주 총 240 l (107 cfu/ml)를 2010년(1년차) 6월 23일, 8월 31일과 2011년(2년차) 4월 18일, 6월 21일에 각각 2번씩 살포하였다. 생장촉진 효과를 측정하고자 20일 간격으로 총 10번 샘플을 채취하였다. 처리군와 대조군 100그루에서 잎을 1,000개씩 임의로 수거하여 엽록소 함량을 엽록소측정기(SPAD 502 PLUS Chlorophyll Meter; Konica, Tokyo, Japan)로 측정하고(Coste 등, 2010; Ling 등, 2011; Uddling 등, 2007), 버니어캘리퍼스(Electronic Digital Micrometer; Chicago Brand, Chicago, IL, USA)를 이용하여 잎의 두께를 측정하였다(Ryu 등, 2011).

엽권정착 PGPB 균주의 혼합제제에 의한 생물적 방제효과를 측정하기 위해서 수거한 벚나무 잎의 진균성갈색무늬구멍병 개수를 평가하였다. 은행나무의 낙엽 정도를 측정하고자 총 구획을 3군데로 나눈 후 전체 개체 중에 낙엽이 든 개체를 측정하여 비율을 계산하였다(은행나무 낙엽수 비율=낙엽이 든 은행나무 개체수/구간 내 전체 은행나무 개체수).

통계분석

본 실험에 이용된 통계분석은 JMPIN4 (SAS Institute, Cary, NC, USA)에서 제공되는 ANOVA (analysis of variance)와 Student's t-test를 이용하여 분석하였다.

결과

분리 균주의 생물학적 활성 분석 및 균주 동정

수목의 엽권에서 내생포자를 만드는 1,056개의 세균 균주를 분리하였다. 분리한 균주의 생물적 활성을 확인하기 위해서 protease, chitinase, lipase 효소 활성을 측정하였다. 분리한 총 1,056균주 중에서 915개 균주가 protease 활성을, 161개 균주가 chitinase 활성을, 27개 균주가 lipase 활성을 보였다(Fig. 1). 길항능력 검정에서 B. cinerea에 대하여 85개 균주, C. graminicola에 대하여 106개 균주가 선발되었다. 3가지 효소와 2가지 진균에 대한 길항능력을 하나라도 보이는 1,022개 균주 중에서, 4개 이상의 복수활성을 가지는 13개 균주와 B. cinereaC. graminicola 모두에 대한 활성만 가지는 1개 균주(8G12)를 포함하여 총 14개 균주를 대상으로 16s rDNA 분석을 통하여 각 균주를 동정하였다. 선발된 14개 균주 중에서 3G7, 4B9, 4C1, 5B4, 5B6, 8D4, 8D9, 8D11, 8E1, 8E11, 8F3, 8G1은 B. subtilis 균주로 판명되었고, 8F6와 8G12는 Paenibacillus polymyxa로 동정되었다(Table 1). 이 중 5B6의 경우 추후의 유전체 분석을 통해서 B. amyloliquefaciens 5B6으로 동정되었다(Kim 등, 2012). 각 균주의 엽권정착능을 확인하기 위하여 각 균주의 107 cfu/ml의 배양액을 고추의 잎에 살포하고 1주일 후에 엽권 내 세균 집락수를 측정하였다(Table 1). 최종적으로 엽권정착 능력이 탁월한 3균주 5B6, 8D4, 8G12를 선발하였다. 8G12균주의 경우 2가지 진균에 대해서만 길항작용을 보이면서 엽권정착능을 보였기에 처리할 PGPB 균주로 최종선발하였다(Table 1).

선발된 엽권정착 PGPB 균주의 고추 포장실험

최종선발된 3개 균주가 포장에서도 효과가 있는지 확인하기 위하여, 모델식물인 고추의 포장실험을 통해 고추의 생장과 병저항성, 수확량을 2010년과 2011년, 2년에 걸쳐 평가하였다. 3개 균주와 혼합처리군을 고추에 살포한 후 식물체의 길이를 측정한 결과, 1년차인 2010년에는 처리 후 20일에 3개 균주(5B6은 62.11 cm, 8D4는 64.15 cm, 8G12는 69.23 cm)는 대조군의 57.24 cm와 비교하였을 때 식물체의 길이가 증가하였다. 1년차 40일에는 5B6이 93.54 cm, 혼합처리군에서 94.79 cm로 대조군의 85.97 cm에 비해 생장촉진 효과를 보였고, 8D4와 8G12는 각각 31.12 cm와 89.71 cm로 대조군보다 길이는 증가하였으나 통계적으로 차이를 보이지 않았다. 1년차 60일에서는 3개 균주와 혼합처리군 모두 대조군의 96.18 cm와 비교하여 각각 5B6에서 123.18 cm, 8D4에서 112.85 cm, 8G12에서 107.55 cm, 혼합처리군에서 105.46 cm로 고추 식물체의 길이가 촉진되는 효과를 보였다. 2년차인 2011년에는 처리 후 20일째 5B6은 63.71 cm와 8D4는 62.32 cm로 대조군의 58.76 cm보다 생장이 촉진되었다. 2년차 60일째에는 3개 균주가 각각 5B6에서 123.16 cm로, 8D4는 119.54 cm로, 8G12는 121.46 cm로 대조군의 115.18 cm와 비교하여 활성을 보였다. 5B6 균주는 1년차 20일에서 8.52%, 40일에서 8.81%, 60일에서 28.08% 증가폭을 보였고, 2년차 20일에서 8.44%, 60일에서 6.93%의 생장 촉진 효과를 보였다(Fig. 2A).

고추의 병 저항성 반응을 확인하기 위해 세균 배양액을 엽면에 살포한 후에 고추에 세균성점무늬병을 일으키는 X. axonopodis pv. vesicatoria를 잎에 접종하여 엽권정착 PGPB 균주에 의한 병저항성 반응을 살펴보았다. 2010년도 고추 점무늬병의 발생정도는 PGPB 처리 후 20일에는 병 감소효과를 보이지 않았지만, 40일째 5B6은 병발생정도 1.72, 8D4는 1.86, 8G12는 1.31로 대조군 2.92 대비 55.0% 병이 감소하였다. 처리 후 60일과 80일은 3균주와 혼합처리군 모두 대조군과 비교해 병발생정도가 감소하였다. 처리 후 60일에는 5B6을 처리했을 때 병발생정도가 1.20, 8D4의 경우 2.04, 8G12 처리 시 1.38, 혼합처리군에서 2.04로 대조군의 3.38과 비교하였을 때 통계적으로 병이 감소하였다. 처리 후 80일에는 5B6에서 1.07로, 대조군의 병발생정도 3.33 대비 68.0%의 병 감소세를, 혼합처리 시 병발생정도가 2.61로 대조군의 3.38과 비교하여 병이 줄어들었다. 2011년도 2년차 포장검정에서는 5B6의 경우 20일에 1.03으로 대조군의 1.56과 비교하여 33.90%의 병 감소를 보였으며, 40일에는 5B6은 1.56, 8D4는 1.32로 대조군의 2.45보다 병이 감소하였다. 60일에는 5B6에서 1.96, 8D4는 2.44, 8G12는 2.53, 혼합처리 시 1.68로 대조군의 병발생정도 3.52보다 최대 46.20%만큼 병이 감소하였고, 처리 후 80일에는 5B6에서 2.02, 8D4에서 2.24, 8G12에서 2.74, 혼합처리군에서 2.45로 최대 49.30% 병이 감소하였다. 처리 후 60일과 80일에서 3균주와 혼합처리군 모두 병 발병이 감소하는 현상을 보였다(Fig. 2B).

마지막으로 고추를 2010년(1년차)과 2011년(2년차) 각각 2차례 수확하여 PGPB의 엽면살포가 고추의 수확량 증대에 어떠한 영향을 미치는지 살펴보았다. 그 결과, 1–2년차 모두 2차 수확 시 고추의 무게를 측정했을 때, 2년차 8G12를 제외한 모든 처리군에서 대조군과 비교하여 더 높은 수확량을 보였다. 1년차 2차 수확 시, 대조군의 1.01 kg과 비교하여, 5B6에서 1.7 kg으로 58.14%, 8D4에서 1.42 kg으로 32.56%, 8G12에서 1.525 kg으로 41.86%, 혼합처리군에서 1.5 kg으로 39.53%의 수확량 증대를 보였고, 2년차 2차 수확 시에도 대조군의 1.05 kg과 비교하여 5B6에서 1.5 kg으로 42.31%, 8D4에서 1.63 kg으로 54.17%, 혼합처리군에서 1.75 kg으로 66.03%의 수확량 증대 효과를 보였다(Fig. 2C).

선발된 엽권정착 PGPB 균주에 의한 가로수의 생장활성 검정. 선발된 PGPB 균주를 대전광역시 유성구 인근에 위치한 은행나무, 벚나무, 이팝나무에 처리하여 고추에서 확인한 엽권정착 PGPB 균주의 활성을 가로수에서도 확인하였다. 가로수의 생장촉진을 확인하기 위해 엽록소 함량과 잎의 두께를 측정하였다. 은행나무의 경우 2010년(1년차) 균주 엽면 처리 20주 후에 67%, 2011년(2년차) 균주 엽면 처리 9주 후 17.41%, 20주차에서 9.71%가 대조군과 비교하여 처리군에서 더 높은 광합성 효율을 보였다. 은행나무 20주차의 경우 은행나무의 낙엽이 지는 20주차 시점에서 상대적으로 더 높은 광합성 효율을 보였다. 벚나무는 1년차 11주차에서 9.47%, 2년차 7주차에서 11.23%, 9주차에서 10.76%로 엽록소 함량이 증가하였다. 이팝나무의 경우에는 1년차 7주차에서 10.46%, 13주차에서 11.56%로 더 높은 엽록소 함량을 보였고 2년차에서는 7주차에서 22.42%로, 17주차에서는 21.25%로 대조군과 비교하여 더 높은 엽록소 함량을 보였다(Fig. 3A).

나뭇잎의 두께는 은행나무의 경우 2010년(1년차) 검정에서 균주 엽면 처리 2주 후에 9.18%, 2011년(2년차) 균주 엽면 처리 9주 후에 4.76%로 대조군과 비교하여 두께가 통계적으로 증가하였다. 벚나무의 잎 두께는 2010년 검정에서 통계적으로 차이를 보이지 않았지만, 2011년에서 균주 엽면 처리 11주 후 8.44%, 15주 후 4.68%로 두께가 증가하였다. 이팝나무의 경우, 1년 균주 엽면 처리 7주 후 19.52%, 13주 후 31.42%로 대조군과 비교하여 두께가 증가하였다. 2년차에 수목 잎의 두께를 측정했을 때 대조군과 다른 처리군에서 통계적으로 큰 차이를 보이지 않았다(Fig. 3B).

선발된 엽권정착 PGPB 균주의 벚나무 진균성갈색무늬구멍병 감소효과와 은행나무 낙엽 생성 촉진 효과

벚나무 진균성갈색무늬구멍병은 진균인 B. jaapii에 의해 자연발생하는 것으로 알려져 있다(Agrios, 1997; Ma 등, 2006). 선발된 PGPB 균주를 수목에 처리한 후에 벚나무상에서 발생하는 진균성갈색무늬구멍병의 수목 잎당 개수의 평균을 측정하였다. 그 결과, 2010년(1년차) 균주 처리 13주 후 67.70%, 19주 후 21.97%로 진균성갈색무늬구멍병 개수가 대조군에 비해 줄어들었다. 또한 2011년(2년차) 균주 처리 15주 후 진균성갈색무늬구멍병 발생이 44.44%, 17주 후 24.14%로 감소하였다(Fig. 4A).

또한 PGPB에 의해 은행나무의 낙엽이 지는 정도를 측정하였다. 혼합제제 처리 17–22주까지의 기간은 은행나무의 낙엽이 지는 시기였기에, 엽권정착세균이 나무 엽권에 정착할 때 나무의 생리적인 활성, 낙엽 형성에 어떤 영향을 미치는지 확인하였다. 또한 광합성 효율 측정에서 2010년(1년차), 2011년(2년차) 모두 균주 처리 20주 후에 엽록소 함량이 증가하였기에 엽록소 함량의 증대와 낙엽생성과의 상관관계를 측정하였다. PGPB 균주를 처리한 후 1년차에서는 17주에서 25.60%, 22주에서 7.14%, 2년차에서는 17주에서 37.10%, 19주에서 14.32%로 대조군과 비교하여 유의미하게 혼합제제 처리군에서 낙엽이 더 많이 발생하는 효과를 확인하였다(Fig. 4B).

고찰

본 연구는 수목의 엽권에 정착하는 내생포자 생성 세균을 분리해 생물학적 활성을 기반으로 분리한 PGPB를 고추와 가로수에 적용하여, 생장촉진효과와 지상부 병을 방제 효과를 확인한 연구이다. 가로수의 엽권에 정착하는 PGPB를 분리한 후, 효소활성 검정을 통해 최종적으로 B. amyloliquefaciens, B. subtilis, P. polymyxa의 3개 종을 PGPB 균주로 선발하였다. 선발된 PGPB 균주를 목본식물의 모델로 이용한 고추에 처리하여 세균성점무늬병 감소와 생장촉진 효과를 확인하였다. 모델식물로 고추에서의 결과를 바탕으로, PGPB를 가로수에 처리했을 때 생장촉진 효과를 확인하였다. 또한 벚나무의 진균성갈색무늬구멍병의 감소와 은행나무의 낙엽이 탈리되는 시기가 앞당겨지는 효과를 확인할 수 있었다. 여기에서는 엽권정착 PGPB를 수목의 엽면에 처리하여 가로수의 종합적이고, 친환경적인 방제 기술을 확립하고자 하였다.

엽권에서 분리한 PGPB를 고추에 살포 시, 고추의 세균성점무늬병과 벚나무의 진균성 진균성갈색무늬구멍병이 대조군에 비해 통계적으로 감소하였다(Fig. 2B, 4A). PGPB에 의한 식물병 방제기작은 경쟁, 길항작용, 식물의 면역을 증진시키는 유도저항성이 알려져 있다(Compant 등, 2005; Glick, 2012; Lee 등, 2015; Saraf 등, 2014). 본 연구에서는 PGPB를 엽면살포하여 엽면에 발생한 식물병의 발생이 감소하는 결과를 설명하기 위해 식물병에 대한 PGPB의 직접적인 길항작용을 살펴보았다. 본 연구에서 분리한 PGPB 중, B. amyloliquefaciens 5B6 균주의 유전체를 분석한 결과, 다른 미생물을 저해하기 위한 항생물질을 생산하는 nonribosomal peptide-synthetase (NRPS), polyketide synthase (PKS) 유전자 클러스터를 확인하였고, 식물 병원균의 세포벽을 용해하는 데 기여하는 surfactin (srf), bacillomycin (bmy), fengycin (fen) 등의 유전자의 존재도 확인하였다(Kim 등, 2012; Koumoutsi 등, 2004). 또한 5B6 균주의 경우 X. axonopodis pv. vesicatoria와 대치배양했을 경우, X. axonopodis pv. vesicatoria의 생장을 억제하였다(data not shown). 이는 PGPB가 생산하는 항생물질에 의해 고추의 세균성점무늬병이 방제된 결과를 설명할 수 있다.

PGPB는 protease, chitinase, lipase와 같은 식물 병원성 진균의 세포벽 구성요소를 분해하는 활성을 갖고 있다(Chernin과 Chet, 2002; Saraf 등, 2014). 벚나무 진균성갈색무늬구멍병은 병원성 진균인 B. jaapii에 의해 발생하는 것으로 알려져 있으며(Agrios, 1997; Ma 등, 2006), PGPB에 의한 효소 활성으로 인하여 벚나무 진균성갈색무늬구멍병이 감소한 것으로 생각된다. 계통상으로 자낭균문(phylum: Ascomycota)에 속하는 C. graminicola, B. cinerea와 벚나무 진균성갈색무늬구멍병(B. jaapii)에 대하여 선발된 PGPB가 길항효과가 있는 것으로 보아, 선발된 PGPB는 자낭균문에 속하는 진균의 공통적인 세포벽 구성성분인 키틴을 분해(chitinase)하거나 세포벽에 부착된 단백질(protease) 혹은 지질이중층(lipase)을 분해함으로써 벚나무 진균성갈색무늬구멍병을 저해한다고 생각할 수 있다(Donmez 등, 2011; Hariprasad 등, 2011; Lee 등, 2012; Maksimov 등, 2011; Neeraja 등, 2010; Planchamp 등, 2015; Tortora 등, 2011).

PGPB는 직접적인 길항작용 외에도, 식물의 유도저항성(induced resistance)을 일으키는 것이 알려져 있다(Choudhary 등, 2007; Compant 등, 2005; Glick, 2012; Glick and Bashan, 1997; Zehnder 등, 2001). 본 실험에서는 선발된 PGPB를 식물체에 엽면살포한 후 엽면에 발생하는 식물병의 발병세를 확인했기 때문에, 선발된 PGPB에 의한 유도저항성 효과를 직접적으로 확인할 수 없었다. 하지만 PGPB인 Bacillus pumilus SE34를 테다소나무(Taeda tree, Pinus taeda L.)에 파종 시 토양에 함께 처리하여, 증진된 ISR에 의해 Cronartium quercuum f. sp. fusiforme가 일으키는 소나무의 혹 녹병을 방제한 사례가 보고되었다. 이를 비추어보았을 때, 선발된 PGPB에 의한 유도저항성 효과도 기대할 수 있다(Bashan과 Holguin, 1998; Compant 등, 2005; Enebak과 Carey, 2000; Eyles 등, 2010; Whipps, 2001).

본 연구에서 선발된 PGPB를 가로수에 처리했을 때 은행나무, 벚나무, 이팝나무에서 엽록소 함량과 잎 두께가 증가하고 고추에서는 길이와 수확량이 증가하였다(Fig. 2, 3). 기존 연구결과에서 기존의 PGPB를 나무에 처리했을 때, 식물 잎의 두께 증가 및 잎의 엽록소의 함량이 증가하였다는 것이 확인되었다(Esitken 등, 2006; Ryu 등, 2011; Stefan 등, 2013). 또한 PGPB 중에서도 엽권에서 분리한 세균이 식물의 생장을 촉진시키고 수확량을 증대시키는 사례도 확인되었다(Dawwam 등, 2013; Esitken 등, 2006; Jacobsen, 1997; Jacobsen 등, 2004; Sahin과 Miller, 1998). 따라서 선발된 PGPB가 수목의 생장에 기여하였음을 확인할 수 있었다.

은행나무에 선발된 PGPB 처리 후 2010년, 2011년 9월에서 11월까지(처리 후 17–22주) 은행나무의 잎의 탈색이 대조군과 비교하여 빨리 일어났다(Fig. 4A). 식물의 낙엽 생성은 일종의 노화반응으로 식물이 외부의 온도와 일조량 변화를 감지하여 광합성 효율이 낮아지면서 잎의 탈색과 탈리 현상이 일어난다(Reid, 1985; Sakamoto 등, 2008b). 식물의 노화는 식물의 예정세포사(programmed cell death)에 의해 일어나는 현상으로 식물 호르몬인 에틸렌과 앱시스산에 의해 조절된다(Gomez-Cadenas 등, 1996; Reid, 1985). 식물 호르몬인 에틸렌에 의한 신호전달기작을 조절하는 ETHYLENE INSENSITIVE2 (EIN2) 유전자가 낙엽 생성에 관여한다는 것을 확인하였다(Bishop, 2009; Kim 등, 2009). PGPB에 의한 식물체의 신호전달 기작을 살펴본 결과, PGPB를 식물체에 처리하였을 때 식물체 내의 자스몬산/에틸렌 관련 신호작용에 영향을 주는 것이 확인되었다(Brock 등, 2013; Fujita 등, 2006; Sakamoto 등, 2008a). 선발된 PGPB 처리 시 에틸렌 신호전달 기작과 관련된 유전자 발현이 증가되는 것으로 미루어 볼 때, PGPB의 작용이 에틸렌을 통해 낙엽의 생성을 촉진하는 것으로 볼 수 있다(Hossain 등, 2011; Pieterse 등, 1998; Wang 등, 2005). PGPB인 Azospirillum을 애기장대와 옥수수에 처리하였을 때 식물체의 노화에 관여하는 다른 식물호르몬인 앱시스산이 축적되었다(Cohen 등, 2008, 2009). 이를 미루어 보았을 때, PGPB에 의한 앱시스산 축적이 수목의 빠른 낙엽을 유도하고, 앱시스산의 축적으로 인해 수목이 겨울철의 저온환경에서도 생존할 수 있게 되는 것으로 생각할 수 있다(Xue-Xuan 등, 2010). 실제로 PGPB인 P. putida를 카놀라에 처리 시 냉해에 견디는 효과가 보고되었다(Sun 등, 1995). 그러므로 PGPB에 의한 에틸렌 관련 유전자 발현량 증가와 앱시스산의 축적은 은행나무에서의 낙엽생성 촉진을 유도하고, 탈색된 잎에 의한 비효율적인 광합성을 제한하고, 저온에 대한 저항성을 획득하게 하여, 겨울철 수목의 생존에 기여할 것이다(Smart, 1994; Xue-Xuan 등, 2010).

본 연구는 수목의 엽권에 정착하는 PGPB를 가로수에 처리함으로써, 기존의 PGPB를 이용한 생물적 방제법을 확장하면서 PGPB의 성공적인 엽권정착을 고려한 가로수의 지속적인 관리방법을 제시하였다. 본 연구에서 실행했던 가로수를 대상으로 한 검정실험에서도 벚나무 병의 방제와 생장촉진효과를 확인할 수 있었기에, 이전의 수목을 대상으로 한 실험 연구결과를 뒷받침하면서 실질적인 가로수 관리 방법을 도입하였다(Ryu 등, 2011). 따라서, 수목의 엽권에서 분리한 PGPB를 수목의 엽권에 정착시킴으로써 PGPB의 엽권정착을 고려한 수목의 새로운 생물적 방제법을 제시하였다.

Enzymatic and antagonistic activities against Botrytis cinerea and Collectotricum graminicola of leaf-associated bacteria isolated from tree species. The Venn diagram presented classification of 1,056 bacterial isolates from leaf surface of tree based on enzymatic activities such as protease, chitinase, and lipase assay and antifungal activities against B. cinerea and C. graminicola. To distinguish endospore-forming leaf-colonizing bacterial isolates from tree phyllosphere, leaf disk samples were soaked in 80°C water bath during 30 minutes.

Antagonistic activities were tested against two fungal pathogens including Botrytis cinerea and Collectotricum graminicola.

Taxonomical identification of 14 selected bacteria were checked using 16s rDNA sequence alignment based on National Center for Biotechnology Information databases.

Leaf colonization means remaining population of each 14 isolates after spraying on 4-week-old pepper leaves. Different letters mean different significance among 14 isolates.
