요 약

고추포장 주변의 모자이크 및 기형의 병징을 보이는 토끼풀에서 CMV를 분리하여 CMV-Tr1으로 명명하였다. 토끼풀로부터 RT-PCR, PCR-restriction fragment length polymorphism 및 외피단백질 유전자의 염기서열 분석을 통해 CMV를 동정하였다. 기주식물 반응결과 N. benthamiana를 포함한 5종의 담배와 오이, 꽈리 및 토마토의 상엽에서 모자이크 병징을 나타내었고, 명아주와 동부에서는 접종엽에 국부괴사 병반을 나타냈다. CMV-Tr1의 외피단백질을 CMV-As와 비교한 결과 99.5%의 상동성을 보였으며, 계통분석 결과 CMV subgroup IB에 속하는 CMV의 한 계통으로 확인되었다.

ABSTRACT

A Cucumber mosaic virus (named CMV-Tr1) isolated from the white clover (Trifolium repens) showing mosaic and malformation that found in a pepper field. Cucumber mosaic virus was identified through confirmation with PT-PCR, PCR-restriction fragment length polymorphism, and sequence analysis of coat protein (CP) gene. CMV-Tr1 mosaic symptom on the upper leaves of five tobacco species including Nicotiana benthamiana, Cucumis sativus, Physalis angulata, and Solanum lycopersicon. In Chenopodium quinoa and Vigna unguiculata the isolate showed local lesions in inoculated leaves. CMV-Tr1 compared with CMV-As in the sequence identity of CP gene. CMV-Tr1 showed 98.9% and 99.5% homologies at nucleotide and amino acid levels, respectively. Phylogenetic analysis of the CP gene indicated that CMV-Tr1 belongs to the CMV subgroup IB base on the CP. To our knowledge, this is the first report of CMV in T. repens in Korea.

서론

Cucumber mosaic virus (CMV)는 오이에서 최초로 보고되었으며(Doolittle, 1916; Jagger, 1916), 85개 과(family) 1,000여 종(species)의 매우 넓은 기주 범위를 가지고 있는 바이러스이다(Choi 등, 1990; Flasinski 등, 1995; Kim 등, 1981; Palukaitis 등, 1992). CMV는 3개의 게놈과 2개의 서브게놈을 가지며(Palukaitis 등, 1992), 다양한 식물에서 발견되는 만큼 계통 또는 분리주(isolate) 간에도 유전적 다양성이 크다(Owen과 Palukaitis, 1988). CMV는 혈청학적 유연관계 및 염기서열의 차이에 따라 subgroup I과 subgroup II로 구분되며(Owen과 Palukaitis, 1988), subgroup I 계통의 CMV의 경우 그룹 내부에서도 염기서열의 차이에 따라 subgroup IA와 subgroup IB로 세분화하기도 한다(Roossinck 등, 1999). 국내의 경우 2007년부터 2011년까지 5년간 주요작물 바이러스병 검정 비율을 분석한 결과 CMV가 가장 높은 검정률을 나타내어 농가에 피해를 끼치는 것으로 보고되었다(Kim 등, 2012).

토끼풀(Trifolium repens)은 우리나라 전체에 걸쳐 자생하는 여러해살이 콩과 식물로 한약재로 쓰이기도 한다. 토끼풀에는 CMV를 포함한 다양한 바이러스가 감염하는 것으로 보고되어(Flasinski 등, 1995), 우리나라에는 Bean yellow mosaic virus, Peanut stunt virus (PSV), Alfalfa mosaic virus, White clover mosaic virus, Red clover vein mosaic virus, Red clover mosaic virus 및 Clover yellow vein virus가 보고되어있다(Jung 등, 2008; Park 등, 2014; Ryu 등, 1986). 본 연구에서는 최근 춘천 근교에 위치한 고추포장 주변에서 모자이크 병징을 보이는 토끼풀로부터 바이러스를 분리하여 검정한 결과 CMV로 동정되어 국내 토끼풀 CMV의 발병을 보고함과 동시에 토끼풀에서 분리된 CMV의 특성을 파악하고자 한다.

기주식물 반응

바이러스에 감염된 것으로 추정되는 토끼풀의 이병주를 채집하여 Nicotiana benthamiana와 N. tabacum cv. Xanthi nc에 접종하였다. 접종 후 육안관찰을 통해 전신감염한 것으로 판단되는 N. tabacum cv. Xanthi nc의 상엽을 채취하여 동부(Vigna unguiculata)에서 단일병반을 분리하였다. N. benthamiana 외 3종의 담배(N. tabacum cv. Xanthi nc, N. glutinosa 및 N. rustica)와 오이(Cucumis sativus cv. Baekdadagi), 토마토(Solanum lycopersicun cv. Prime Alexander) 및 동부 등에 접종한 결과 오이의 상엽에 모자이크 병징을 나타내었고, 동부의 접종엽에서 CMV의 특징적인 병징인 괴사 반점이 관찰되었다(Table 1). 이것은 기존에 토끼풀에서 보고되었던 다른 바이러스종에서는 나타나지 않는 병징으로 Cucumovirus속의 다른 바이러스인 PSV의 특성과도 구분되는 CMV의 한 계통으로 추정되었다.

RT-PCR 및 제한효소지도

기주식물 반응을 통해 토끼풀에 감염한 바이러스가 CMV의 계통으로 추정되어 CMV의 외피단백질을 검정하는 특이 프라이머(CMV CP-5: 5’-ATGGACAAATCTGAATCAACC-3’, CMV CP-3: 5’-TCAGACTGGGAGCACTCCAGA-3’)를 이용하여 대조구로 서울여대 바이러스은행에서 분양받은 CMV subgroup IA (CMV-Fny)와 subgroup IB (CMV-As)와 함께 RT-PCR을 수행하였다. 증폭된 PCR 산물을 1.2% agarose gel에서 전기영동으로 확인한 결과, 대조구로 사용된 CMV-Fny 및 CMV-As의 증폭산물과 크기가 유사함을 확인할 수 있었다(Fig. 1). 이를 통해 토끼풀로부터 분리한 바이러스가 CMV의 한 계통으로 확인되어 CMV-Tr1으로 명명하여 이하 실험을 수행하였다. CMV-Tr1의 특성과 계통 분석 위해 EcoRI, HindII, SacI 등 3가지 제한효소를 사용하여 제한효소지도 분석을 실시하였다(Fig. 2). 그 결과 CMV-Tr1은 EcoRI에서만 PCR 산물이 절단되는 것을 확인하였으며, 이것은 CMV subgroup IB 계통의 바이러스들이 갖는 특성과 일치하는 것으로 대조구인 CMV-Fny와 CMV-As를 통해 확인할 수 있었다.

염기서열 및 계통 분석

CMV-Tr1의 RNA3에 존재하는 외피단백질(coat protein)의 염기서열을 결정하여 계통분석(MEGA6; Arizona State University, Tempe, AZ, USA)을 실시한 결과 subgroup IB 계통의 바이러스와 가장 유사한 특성을 나타냈다(Fig. 3).

토끼풀에 감염하는 바이러스로 아직까지 국내에서는 보고되지 않았던 CMV-Tr1의 분리를 통해 국내에도 토끼풀에 CMV가 감염하고 있음을 확인할 수 있었다. 또한 여러해살이풀인 토끼풀에서 CMV와 함께 같은 Cucumovirus속의 PSV가 자연 감염으로 인한 월동기주 역할을 할 것으로 예상되었다(Jung 등, 2008; Mink 등, 1969). 따라서 토끼풀에서 분리된 CMV-Tr1을 다른 여러 식물에 대한 기주 실험을 실시하여 다양한 과의 작물에서의 특성을 분석할 필요가 있다.
