토마토황화잎말림바이러스병에 대한 저항성 품종과 항바이러스 활성 물질 3종의 효과 검증

요 약

최근에는 작물의 유도 저항성을 이용한 항바이러스제 개발에 관한 많은 연구가 수행되고 있으나 실제 농업현장에 널리 보급되지 못하고 있는 실정이다. 본 실험은 시설토마토 재배현장에 외생 살리실산, 키토산, 유제놀 처리에 따른 토마토황화잎말림바이러스(Tomato yellow leaf curl virus, TYLCV) 감염억제효과를 검증하고자 수행되었다. 실내검정에서 TYLCV에 감수성 품종인 ‘슈퍼도태랑’은 항바이러스제 처리 후 TYLCV에 감염된 지 12일 후 VP (virus infected control plants)에서 바이러스 증상이 나타나기 시작했다. 접종 32일 후 TYLCV 발병도는 VP에서 98.8%였고, SAT (2 mM salicylic acid)+VP, EGT (200 μ g/ml eugenol)+VP에서는 각각 98.8%, 98.7%로 발병도가 높았으나, CHT (0.1% chitosan)+VP는 85.7%로 다른 처리들과 통계적으로 유의한 차이를 보였다. 그러나 TYLCV 농도는 CHT+VP에서 OD 값이 0.3으로 오히려 가장 높게 나타났으며, 토마토의 초장, 지상부 및 지하부 생체중에서도 뚜렷한 효과를 보이지 않았다. 여름작형 시설재배지에서 도태랑 계열의 토마토품종 ‘도태랑솔라’를 사용하여 항바이러스 3종의 효과를 조사한 결과, 수확기에 모든 처리구에서 100.0%에 가까운 TYLCV 감염률은 나타냈으며, 수확량에도 처리간의 통계적 유의차가 인정되지는 않았다. 이와 대조적으로 Ty-1Ty-3a 유전자를 보유한 TYLCV에 내병성 품종인 ‘ TY자이언츠’는 저항성 유묘검정의 전 조사기간 동안 바이러스 증상이 전혀 관찰되지 않았고, 식물체내 바이러스 농도도 무접종 수준이었다. 본 실험 결과 ‘ TY자이언츠’ 품종은 TYLCV 발생이 만연한 지역 및 재배시기에 감수성 품종을 대체할 수 있을 것으로 생각된다. 반면, 저항성 유도물질인 살리실산, 유제놀, 키토산의 항바이러스 효과는 입증되지 않았기 때문에, 아직 시설토마토 재배현장에 적용하기는 어려울 것으로 판단된다.

ABSTRACT

Recently, several in vitro studies have reported antiviral activity of agents of systemic acquired resistance against plant virus infection, but the approach has not been applied in a wide range of agricultural fields. The objective of this study was to evaluate the inhibitory effect of the exogenous application of salicylic acid (SA), chitosan (CH), or eugenol (EG) in tomato yellow leaf curl virus (TYLCV) infection of greenhouse-grown tomato plants. In vitro, the initial time of symptom was observed in TYLCV-infected plants (VP) of the resistant cultivar ‘Superdotaerang’ at 12 days post inoculation (dpi) after application of antiviral agents. At 32 dpi, the disease rate of TYLCV in the CHT+VP (0.1% chitosan and virus infected control) treated plants was 87.5%, lower than that of the other treatment. However, the virus content in the CHT+VP treated plants was higher than those of the other treatments, and SA, EG, and CH did not show significant effect on plant height or shoot and root fresh weight. Our results from summer-cultivated greenhouse-grown tomatoes show that none of the tested agents had an inhibitory activity on viral infection or yield of tomato ‘Dotaerangsola'cultivar. In contrast, all treated ‘TY Giants’ cultivars that possessed TYLCV resistance genes Ty-1 and Ty-3a did not show typical symptoms and the virus content was remarkably lower than those in the TYLCV treated plants in ‘Superdotaerang’. The results of this research indicated that the planting of resistant tomato cultivars was effective method instead of using SA, EG, and CH (known as resistance-inducing factors for control) of TYLCV in the field.

서 론

토마토황화잎말림바이러스(Tomato yellow leaf curl virus, TYLCV)는 GeminiviridaeBegomovirus속에 속하는 single-stranded DNA 바이러스로(Brown 등, 2015), 최소 11개의 계통이 보고되어 있는데(Fauquet 등, 2008), 1964년 이스라엘에서 처음 분리된 TYLCV 분리주(TYLCV-IL)는 전 세계적으로 발생되고 있으며 심각한 경제적 피해를 일으키고 있다(Cohen과 Harpaz, 1964; Cohen과 Lapidot, 2007; Lefeuvre 등, 2010; Moriones 등, 2011).
TYLCV는 토마토, 감자, 담배 등 Solanum 속에 다양한 작물을 감염시키며(Polston과 Anderson, 1997), 국내에서는 가는잎한련초, 큰메꽃, 쇠별꽃 등 9과 13종의 자연 기주잡초를 감염시킬 수 있는 것으로 보고되었다(Kim 등, 2012). 이 바이러스에 감염된 토마토는 잎 가장자리가 황화되고 잎이 말리는 증상이 나타나며 병의 진전에 따라 식물 전체가 위축되고 총생되어 일반적으로 28-92%의 수확량 손실을 일으키지만, 생육 초기 단계에서 감염되면 수확을 거의 할 수 없어 최대 100% 수량감소를 초래한다(Moriones과 Navas-Castillo, 2000; Nakhla과 Max-well, 1998).
TYLCV는 담배가루이에 의해 영속 전염되기 때문에 기존 바이러스 예방법은 매개충 방제에 집중되어있었다. 그러나 국내에 우점하고 있는 담배가루이 MED (Mediterranean)는 네오니코티노이드 계통과 insect growth regulator (IGR) 계통인 피리프록시펜에 저항성을 나타내는 것으로 보고되었다(Kim 등, 2008; Lee 등, 2000, 2012). 이렇듯 약제저항성 발달은 농약의 고농도 과량 살포를 유발해 농업환경을 오염시키는 등의 문제를 야기할 수 있다. 따라서, TYLCV 피해를 줄이는 환경 친화적인 방법으로 내병성 품종 개발과 작물의 저항성 유도물질을 이용하는 연구가 활발하게 진행되고 있다. 현재까지 내병성 토마토 육종에 사용되는 TYLCV 내성 유전자는 최대 6개(Ty-1, Ty-2, Ty-3, Ty-4, Ty-5, Ty-6)이며 S. pimpinellifolium, S. peruvianum, S. chilense, S. habrochaitesS. cheesmaniae 등 모두 야생토마토에서 유래되었다(Butterbach 등, 2014; Ji 등, 2007; Scott, 2007). 그 중 Ty-1은 상업용 품종에 가장 많이 사용되는 유전자로 S. chilense 계통인 ‘ LA1969’ (Zamir 등, 1994)에서 보고되었고, 같은 야생종의 ‘ LA1932’ 계통에서 Ty-3, S. habrochaites 계통인 ‘ B6013’에서 Ty-2 (Ji 등, 2007; Verlaan 등, 2013)가 보고되었다. TYLCV 내병성 토마토 유전자형은 식물체내에서 바이러스 복제를 완전히 억제하지 못하고 전신감염되어 경미한 증상으로 발현되기도 하나 감수성 유전자형에 비해 낮은 수준의 바이러스 축적으로 수확량 손실을 감소시킨다(Lapidot 등, 2001; Legarrea 등, 2015).
전신획득저항성(systemic acquired resistance, SAR)은 병원체 침입 등에 의해 피해를 입었을 때 그 피해를 입은 부위로부터 signal이 식물 전체로 퍼져 식물체 전체에 저항성이 생기게 되는 현상을 일컫는다(Ryals 등, 1996; Yalpani 등, 1993). 담배, 애기장대풀 등의 식물에서 밝혀진 바에 의하면, SAR 발현에는 식물체내의 발병기전 단백질(pathogenesis-related protein, PR protein)의 축적이 유도되는데, 이러한 PR 유전자 발현의 활성화 및 SAR 반응은 살리실산(salicylic acid, SA)이 매개하는 것으로 알려져 있다. 외생 SA 처리는 식물의 생물적, 비생물적 환경변화에 대한 저항성을 증대시킬 뿐만 아니라(Esmailzadeh 등, 2008; Javaheri 등, 2012; Khan 등, 2015, 2019; Koo 등, 2020), 항바이러스 활성도 향상시키는 것으로 보고되었다(Calil 과 Fontes, 2017; Li 등, 2017, 2018). 토마토 잎에 SA를 0.1 mM 의 농도로 처리하였을 때 오이모자이크바이러스(Cucumber mosaic virus, CMV), 담배모자이크바이러스(Tobacco mosaic virus, TMV) 및 순무주름바이러스(Turnip crinkle virus, TCV)의 복제를 억제하였고 담배의 CMV 감염을 지연시켰다(Faoro과 Gozzo, 2015; Shang 등, 2011). SA가 바이러스 복제, 세포 간 이동 및 장거리 수송을 포함한 바이러스 감염과정의 세 가지 주요 단계에서 간섭 할 수 있다는 것이 다른 여러 연구를 통해 입증되었다(Tahmasebi 등, 2011). 외생 SA 외에도 다양한 식물추출물이 항바이러스제로 사용되고 있으며, 그 중 계피오일의 주요 활성 성분인 유제놀(eugenol, EG)은 산화질소(nitric oxide, NO)와 SA 축적을 높여 토마토 식물에서 TYLCV에 대한 저항성을 유도하는 것으로 알려져 있다(Sun 등, 2016; Wang과 Fan, 2014). Tsai 등(2019)의 실험에서 토마토에 200 μ g/ml EG 전처리는 TYLCV 발생률을 60%까지 감소시켰다. 키토산(chitosan, CH)은 N-acetyl-D-glucosamine이 β-(1→4) glycosidic linage 결합으로 연결된 천연 고분자 화합물로 새우, 게, 곤충 등 갑각류의 외골격과 진균의 세포벽의 구성 요소인 키틴을 탈아세틸화하여 얻어진다(Iriti과 Varoni, 2015). 키틴과 CH의 파편은 미생물 감염에 대한 반응으로 식물에서 다양한 방어 반응을 유도하는 것으로 알려져 있으며, 주로 자스모네이트(jasmonate) 생합성을 유도하여 저항성 유전자 발현을 조절한다(Doares 등, 1995). 이전 연구에 따르면, 콩잎에 0.1% CH 전처리는 알팔파모자이크바이러스(Alfalfa mosaic virus, AMV)에 의한 국부병반 생성을 차단하였고(Pospieszny 등, 1991), 동일한 농도로 CH를 토마토에 처리하고 감자걀쭉바이로이드(Potato spindle tuber viroid, PSTVd)를 접종한 실험에서도 이와 유사한 억제효과를 나타내었다(Pospiezny, 1997). Mishra 등(2014)에서는 CH를 Pseudomonas 속의 근권미생물과 혼합처리하였을 때 토마토의 생육을 증진시키고, Tomato leaf curl virus (ToLCV) 질병의 중증도를 80-90% 감소시키는 효과를 확인하였다. 이렇듯 다수의 연구를 통해 TYLCV에 대한 외생 SA, EG, CH의 방제효과가 입증 되었음에도 불구하고 아직까지 실제포장에서 바이러스병 방제 소재로 적용된 사례가 부족하여 널리 사용되지 못하고 있다. 따라서, 본 연구에서는 토마토에 항바이러스제 3종을 처리하였을 때 TYLCV에 대한 감염 억제 효과 및 생육 효과를 실내검정하고, 시설토마토 재배 포장에서 TYLCV에 대한 방제효과를 비교하는 한편, 경기 광주지역에서 주로 재배되고 있는 완숙토마토 품종의 TYLCV 내성 유전자 보유여부를 확인하고, 발병정도를 조사하여 영농현장에서 농업자재 및 토마토 품종 선택을 위한 기초자료로 활용하고자 하였다.

재료및 방법

식물재료 및 바이러스 접종. 실내 실험에 사용한 토마토 품종은 ‘슈퍼도태랑’ (코레곤)과 ‘ TY자이언츠’ (미푸코) 2종으로 1.0% NaOCl에 1분 동안 표면살균한 종자를 멸균한 피트모스와 질석(1:1, w/w) 혼합물에 파종하였다. 파종한 종자는 25°C 암조건에서 발아시켜 자엽이 전개되었을 때, Hoagland 배양액(MBcell, Seoul, Korea)을 주기적으로 관주하여 2주간 생장상에서 주간 26°C, 야간 23°C, 광도 185 μ mol/m2/s 조건으로 육묘하였다. 바이러스 접종원은 국내 발생 TYLCV-IL에 대하여 선행연구(Kil 등, 2014)에서 사용한 감염성 클론을 사용하였으며, Kil 등(2014)의 방법에 따라 배양하였다. 형질전환된 Agrobacterium tumefaciens를 3종의 항생제(50 μ g/ml 의 kanamycin, 50 μg/ml의 rifampicin, 10 μg/ml의 genta-mycin)를 첨가한 Luria-Bertani 배지에서 28°C, 24시간 진탕배양하였다. 접종은 2-3엽기에 토마토 줄기 정단부를 곤충핀을 이용하여 10회 가량 반복하여 찔러준 후, 배양액 500 μ l를 찔러준 부위에 일회용 스포이드를 이용하여 조심스럽게 떨어뜨리면서 흡수되도록 하였다. 접종한 식물체는 주야간 온도가 26°C와 23°C로 유지되는 생장상(광/암, 16 hr/8 hr; 습도, 50% 이상)에서 재배하였다.
항바이러스제 실내검정. 토마토 ‘슈퍼도태랑’ 품종을 파종하여 3-4 엽기된 유묘에 SA (2-hydroxybenzoic acid 99%, Sigma-Aldrich, St. Louis, MO, USA)과 EG (2-methoxy-4-(2-propenyl)phenol 99%, Sigma-Aldrich), CH (poly(D-glucos-amine) 500-190 kDa, Sigma-Aldrich)을 흘러내릴 정도로 충분히 분무처리하고, 24시간 뒤 TYLCV 감염 클론을 접종(virus infected control, VP)하였다. SA과 EG, CH의 엽면살포 농도는 이전문헌을 참고하여 각각 2 mM SA, 200 μ g/ml EG, 0.1% CH로 선정하였다(Li 등, 2019; Rendina 등, 2019; Sun 등, 2016). 실험은 SAT+VP, EGT+VP, CHT+VP, VP 및 무접종(CP)으로 총 5개 처리구로 구성하였고 식물 생장상에서 난괴법으로 배치하였으며, 처리당 토마토를 12주씩 3반복으로 실험을 진행하였다. 항바이러스제 3종 처리 및 TYLCV 감염이 토마토 생육에 미치는 영향을 조사하기 위해 처리 50일 후 각 처리구별 초장과 지상부 및 지하부 생체중을 측정하였다. TYLCV에 대한 항바이러스효과 조사는 TYLCV 접종 18일 및 32일 후, 처리별 토마토에 TYLCV 발병도를 조사하였고, double antibody sandwich enzyme linked immunosorbent assay (DAS-ELISA) 방법으로 바이러스 농도를 비교하였다. 신초 잎을 0.1 g씩 무작위로 채취한 후 추출액 1 ml을 첨가하고, zirconia beads를 이용하여 마쇄한 후 원심분리한 상징액을 사용하였다. TYLCV 항체 검출용으로 시판중인 TYLCV complete kit (Bioreba, Reinach, Switzerland)를 사용하였으며, 제조사에서 권장하는 방법에 따라 실시하였다. 바이러스 측정은 ELISA Microplate Reader (SpectraMax 190, Molecular Devices, Sunnyvale, CA, USA)를 사용하여 450 nm 의 파장에서 흡광도를 측정하였다.
항바이러스제 포장검정. 항바이러스제 3종의 농가실증 시험은 경기도 광주시 퇴촌면 정지리에 위치한 농가 하우스에서 실시되었으며 육묘장에서 구입한 ‘도태랑솔라’ (코레곤) 품종의 토마토를 2020년 7월 3일에 정식하여 6개월 동안 실험을 수행하였다. 실험에 사용된 처리구는 SAT, EGT, CHT, 무처리(NT)로 총 4개로 구성하였고, 정식 직후 2 mM SA, 200 μ g/ml EG, 0.1% CH를 1주일 간격으로 수확 직전까지 총 8회 지상부를 충분히 적실만큼 살포하였다. TYLCV 감염률 조사는 정식 30일 뒤와 90일 뒤에 각각 실시하였으며, 병징의 유무와 상관없이 토마토 신초 잎을 무작위로 채집하여 polymerase chain reaction (PCR)을 실시하였다. 채집한 시료는 자동핵산추출장비인 magLEAD 12gC (Precision System Science Co., Matsudo, Japan)를 사용하여 total DNA/RNA를 분리하였고, TYLCV 검출을 위해 Lee 등(2013)에 의해 보고된 특이 프라이머 TYLCV-M1F (CACGATT-TAATTAGGGATCTTATATCTG)와 TYLCV-M1R (ATCAGGGCTTC-GATACATTC)를 이용하여 PCR을 수행하였다. 토마토의 수확은 정식 55일 후부터 시작하였고, 1화방 조기수량과 5화방까지 전체 수량을 측정하였으며, 통계처리 분석을 통하여 처리별 수량성 차이를 분석하였다.
바이러스 저항성 검정. 감염성클론 TYLCV-IL을 사용하여 토마토 품종 ‘슈퍼도태랑’ 및 ‘ TY자이언츠’에 접종하였으며, 접종한 식물체는 주야간 온도가 26°C와 23°C로 유지되는 생장상(광/암, 16 hr/8 hr; 습도, 50% 이상)에서 30일 동안 재배하면서 병징을 조사하였다. 바이러스 접종은 품종 당 10주씩 3반복으로 실시하였다. 바이러스 조사는 Choi 등(2013)의 방법을 변형하여 증상 없음(0), 약한 증상(1), 황화증상(2), 위축과 황화 잎말림(3)의 4단계로 구분하고 다음과 같이 발병도(발병도(%)=[Σ(발병수×발병계수)/3×개체수]×100)로 환산하였다.
TYLCV 내병성 유전자는 Ty1/3과 연관된 cleaved amplified polymorphic sequences (CAPS) 마커 (‘ TY1-SspI’)와 내성 유전자 Ty-2, Ty-3 또는 Ty-3a와 연관된 sequence characterized amplified region (SCAR) 마커 (‘ TO302’, ‘ P6-25’)를 이용하여 검정하였다(Table 1). Genomic DNA는 토마토 본 잎 5-7엽기에 자동핵산추출시스템인 magLEAD 12gC를 사용하여 매뉴얼에 따라 추출하였다. SCAR 마커 PCR 수행을 위해 gDNA 2 μ l에 SCAR primer 각각 20 pM과 PCR 반응액(0.025 U KOD-FX polymerase, 0.2 mM dNTP; TOYOBO, Osaka, Japan) 20 μ l 를 섞고 94°C에서 10분 pre-denature를 수행한 후, denature 94°C 30초, annealing 60°C 30초, extension 72°C 2분으로 총 34 cycle로 반응시킨 후 last extension 72°C에서 5분간 진행시켰다. CAPS 마커 분석은 증폭된 PCR 산물 5 μ l를 이용하여 총 10 μ l의 반응액에 0.5 U의 제한효소를 처리한 후 1× TBE buffer (Bioneer, Daejeon, Korea)가 첨가된 2% agarose gel을 이용하여 75 V에서 35분간 전기영동을 실시하여 UV illumination상에서 관찰하였다.

통계분석. 수집된 자료의 통계분석은 SPSS프로그램(IBM SPSS Statistics ver. 26.0, IBM Corp., Armonk, NY, USA)를 이용하여 분산분석(one-way analysis of variance)후 Duncan 다중 범위검정(Duncan multiple range test, 5%)을 이용하여 유의성의 차이를 검정하였다.

결과및 고찰

항바이러스제 실내검정. 항바이러스제 3종의 엽면살포 후 TYLCV 접종을 수행한 결과(Table 2), TYLCV에 감염된 지 12일 후 VP에서 생장점이 황화되고 잎이 말리는 증상이 나타나기 시작했고, 접종 18일 후부터 CP를 제외한 모든 처리구에서 TYLCV 증상이 발현되었다. 처리 후 50일째 바이러스를 접종한 모든 처리구의 초장은 CP에 비해 낮았고, 지상부 생체중의 경우 EGT+VP은 8.6 g, CHT+VP은 8.7 g, SAT+VP은 9.1 g, VP은 10.0 g 순으로 높았다. 본 연구결과 TYLCV의 감염으로 인해 토마토의 초장은 감소했지만 오히려 지상부 생체중은 증가한 것으로 미루어볼 때 식물 체내에 침입한 바이러스의 작용이 토마토의 생리, 생장반응을 비정상적으로 변화시킨 것으로 판단된다. Seo 등(2018)에서 TYLCV 감염으로 발현되는 위축, 왜화 등의 병징에는 셀룰로오스 생합성 경로와 관련된 다양한 유전자가 관여하는 것으로 보고되었고, TYLCV의 C4 단백질이 여러 식물발달 관련 유전자의 전사 경로를 방해하여 잎의 말림증상을 유도하는 것으로 밝혀졌다(Corrales‐ Gutierrez 등, 2020). 뿐만 아니라 TYLCV는 식물 체내 수분 손실을 막기 위해 증산율과 기공전도도를 감소시켰으며, 일부 탄수화물과 아미노산 등의 삼투 보호제를 뿌리로 재분배하여 R/S (root/shoot) 비율을 증가시켰다(Mishra 등, 2022; Shteinberg 등, 2021).

따라서, 본 연구결과 바이러스 접종 처리구의 초장 감소는 TYLCV의 전형적인 병징 발현에 기인한 결과이며, 생체중의 증가 또한 TYLCV 감염에 따른 체내 삼투보호제의 축적 때문일 것으로 추측된다. 반면, VP와 비교하여 SAT+VP, CHT+VP, EGT+VP 의 지상부 및 지하부 생체중이 상대적으로 낮았는데, 이는 항바이러스 3종의 처리가 감염초기 식물체내 SAR 유도작용으로 자체 방어기작을 활성화시켜으나, 식물 생육은 억제시킨 것으로 생각된다.
TYLCV가 침입 후 성공적인 감염을 위해 식물의 과민성세포사멸반응(hypersensitive cell death)을 유도하지 않으며, 기주 식물의 샤페론 열충격단백질 90 (heat shock protein 90)을 비활성화하고, 열충격전사인자(heat shock transcription factor)를 억제하는 등의 식물 방어 반응을 회피하는 것으로 알려져 있다(Anfoka 등, 2016). 반면, SA에 의해 매개된 방어 반응의 활성화는 종종 식물의 성장을 억제하는 것으로 알려져 있는데(Huot 등, 2014; Karasov 등, 2017), 이전에 몇몇 연구에서는 외생 SA의 처리가 일부 식물 종에서 처리 농도에 따라 영양성장을 감소시켰다(Rivas-San Vicente와 Plasencia, 2011). Matricaria chamomilla에 250 μ M SA 처리는 지상부 및 지하부의 생장을 억제시켰고(Kovácik 등, 2009), Arabidopsis에서도 50 μ M 농도의 SA 처리가 뿌리 생장을 억제시킨다는 보고가 있었다(Pasternak 등, 2019).
처리별 TYLCV 발병도 조사 결과 접종 18일 후 VP에서는 78.6%였고, SAT+VP, CHT+VP, EGT+VP에서는 각각 56.8%, 59.5%, 55.1%로 항바이러스제 3종의 처리구에서 유의하게 낮게 관찰되었다. 접종 32일 후에는 VP에서 98.8%였고, SAT+VP, EGT+VP에서는 각각 98.8%, 98.7%로 발병도가 높았으나, CHT+VP는 87.5%로 다른 처리들과 통계적으로 유의한 차이를 보였다(Table 2, Fig. 1). 본 실험 결과와 비슷하게 토마토 감수성 품종 ‘ Jinpeng-1’에서 SA 처리가 TYLCV 접종 후 10일까지 대조구와 비교하여 체내 바이러스 함량을 유의하게 감소시켰는데, 이는 2 mM 농도의 SA 처리가 체내 활성산소종(reactive oxygen species) 생성을 억제하고 병 저항성 관련 효소를 조절함으로써 TYLCV에 대한 저항성을 증대시켰기 때문으로 확인되었다(Li 등, 2019). EG 처리는 NO와 SA의 함량을 축적하여 TYLCV에 대한 저항성을 향상시킨다고 알려져 있다(Sun 등, 2016; Wang과 Fan, 2014). Tsai 등(2019)에서는 200 μ g/ml EG 전처리가 Tomato leaf curl New Delhi virus DNA A와 베타위성 DNA (DNA B)의 축적 속도를 감소시켰으나, 접종 15일 후에는 DNA B의 농도가 대조구와 비슷한 수준으로 증가했다고 보고하였다.

DAS-ELISA 결과를 비교해 보면, 접종 18일 후 발병도는 VP에 비해 항바이러스 3종 처리구에서 발병도가 유의하게 낮게 나타났고 TYLCV 농도는 SAT+VP의 OD값이 0.2로 가장 낮았으나 처리간 유의차는 인정되지 않았다. 접종 32일 후 CHT+VP에서 발병도가 87.50%로 다른 처리구에 비해 낮게 나타났으나, TYLCV 농도는 OD값이 0.3으로 오히려 가장 높게 나타났다(Table 2, Fig. 2). 따라서, 본 연구의 결과와 Lapidot 등(1997)의 결과로 미루어 보아 식물체의 바이러스 농도가 발병도와 언제나 일치하는 것은 아니라는 것을 확인했으며, 감염 후 바이러스 병징의 정도에 따라 체내 바이러스 증식농도에 변화가 있을 것으로 추정되었다.

시설토마토 재배지에서 TYLCV 감염억제 효과 검정. 항바이러스제 3종 처리에 따른 토마토 TYLCV 감염률과 수확량을 비교해 보면, 토마토 정식 후 30일째인 제 1화방 착과 비대기에 TYLCV 감염률은 NT 1.2%, SAT 0.8%, CHT 0.8%, EGT 0.5% 순으로 낮았고, 1화방 누적 수확량은 NT 963 kg/10a, CHT 1,079 kg/10a, SAT 1,102 kg/10a, EGT 1,116 kg/10a 순으로 많았으나 처리 간 차이는 없었다. 제 3화방 수확기에는 CHT 100.0%, SAT 100.0%, EGT 98.3%, NT 96.7% 순으로 낮게 나타났으며, 1화방에서 5화방까지 처리별 전체 수확량은 SAT 3,460 kg/10a, CHT 3,640 kg/10a, NT 4,235 kg/10a, EGT 4,305 kg/10a 순으로 많았으나 통계적으로 유의차가 인정되지 않았다(Table 3). Abd El-Gawad와 Bondok (2015)에 보고에 의하면 2 mM SA와 0.1% CH의 혼합처리가 TMV 감염을 억제하고 단독처리에 비해 토마토 수량을 증가시키는 효과가 있었고, CH과 Pseudomonas sp. 혼합처리가 ToLCV 발병도를 감소시킬 뿐 아니라 식물 성장을 촉진하고 토마토의 주당 수량을 증가시키는 등 미생물과 식물 병 저항성 유도물질의 조합처리에 따른 상승 효과가 보고된 바 있어(Mishra 등, 2014), 항바이러스 효과가 인정되는 물질들의 조합에 대해서도 추가 연구가 필요할 것으로 생각된다.

최근 연구에 따르면 TYLCV 증상은 봄 작형에 비해 겨울 작형에 경미하게 나타나며(Rashid 등, 2008; Singh 등, 2015), 온도 상승에 따라 TYLCV 발생률이 증가하는 것으로 보고되었다(Iftikhar 등, 2021). 이는 궁극적으로 고온으로 인해 매개충인 담배가루이 발생 증가에 기인한 것으로 분석되었다(Zeshan 등, 2016). 따라서 본 연구 결과, 생육초기 TYLCV 감염수준이 0.5-1.2%였으나 수확기에 접어들면서 감염률이 96.7-100.0%로 급증하였던 것은 담배가루이의 밀도가 높은 여름재배 작형에 본 연구가 수행되었기 때문으로 판단된다. 실험에 사용된 항바이러스 3종의 효과가 감염초기에만 일부 인정되었던 이전 연구결과로 미루어 볼 때, 전 생육기간 동안 바이러스의 감염위험에 노출되어있는 토마토 재배현장에서는 아직 적용하기는 어려울 것으로 생각된다.
토마토 시험품종의 TYLCV 내병성평가. 시험품종 ‘슈퍼도태랑’에서는 내성 유전자 마커가 발견되지 않았으며, 발병률도 94.4%로 높았다. 반면, TYLCV 내병성으로 알려진 대비품종 ‘ TY 자이언츠’는 Ty1/3 유전자좌에서의 이형접합이 발견되었고 Ty-3a 유전자 존재가 확인되었으며, 발병률이 0.0%로 매우 낮았다(Table 4, Fig. 3). Verlaan 등(2013)에서 TYLCV에 대한 내성 유전자 Ty-1Ty-3 또는 Ty-3a가 6번 염색체에 존재하며, 두 유전자가 동일한 유전자좌의 대립유전자임이 입증됨에 따라 ‘ TY자이언츠’ 품종의 Ty-1Ty-3 유전자는 부모 중 한쪽으로부터 유래되었을 것으로 판단된다. 또한 Ty-1 내성을 담당하는 유전자가 DFDGD-class RNA-dependent RNA polymerase로 확인되었고, TYLCV의 DNA 복제 및 메틸화에 영향을 미치는 것으로 보고되었다(Butterbach 등, 2014). 본 연구에서 활용한 Ty1/3 유전자좌에 대한 CAPS 마커는 Ty-1Ty-3 또는 Ty-3a 내성 유전자를 포함하고 있으며 이 유전자들를 보유한 다수의 유럽품종에서 TYLCV 발병률이 3.3% 이하로 매우 낮게 나타났다(Kim 등, 2013).

따라서, 본 연구에 활용한 CAPS 마커를 이용하여 TYLCV 내성 유전자 보유 여부를 확인한 후에 품종을 선택하는 것이 바이러스병의 대발생을 예방하고, 토마토의 안정생산에 도움이 될 것으로 생각된다.
