아욱에서 분리한 Malva Vein Clearing Virus 분리주의 특성

ABSTRACT

In September 2017, vein clearing and yellowing symptoms resembling those caused by viruses were observed on leaves of Malva verticillata in Chungnam, Korea. Nucleic acids were extracted from leaves of five symptomatic plants and tested by reverse transcription polymerase chain reaction using four virus specific primer pairs including malva vein clearing virus (MVCV). Amplicons of the expected size (600 bp) were obtained from total RNA of all samples using the MVCV-specific primers. To confirm the presence of MVCV in symptomatic plants, the DNA fragments from three samples were purified, and directly sequenced. BLAST analysis revealed that it shared the highest nucleotide identity (99%) with a MVCV isolate from tomato (Mexico). The virus isolates obtained from the third re-inoculated Chenopodium was designated as Cm1-5. Tissue from Cm1, Cm3, and Cm5 isolates was mechanically sap inoculated into 23 indicator plants. Cm3 isolate induced chlorotic local and mosaic symptoms in Althaea rosea. Phylogenetic analysis based on coat protein gene of 19 MVCV isolates from 6 different countries and plant species, did not correlated with either the geographical origin of the isolates, or pathogenicity. To our knowledge, this study first reports the natural occurrence of MVCV on M. verticillata in Korea and characterization of three Korean isolates of MVCV.

아욱엽맥투명바이러스(Malva vein clearing virus, MVCV)는 Potyvirus 속(genus)에 속하는 바이러스로, 진딧물 및 즙액을 통해 전염되는 것으로 알려져 있다(Horváth 등, 1979). 1956년 독일지역의 당아욱(Malva sylvestris, mallow)에서 처음 보고되었으며(Hein, 1956), 그 후 유럽(스페인, 이탈리아 등), 미국(캘 리포니아), 이란, 중국 등에서 아욱속(Malva sp., M. assurgentiflora, M. nicaensis, M. paraviflora, M. rotundifolia), 라바테라속 (Lavatera cretica, L. trimestris), 접시꽃(Alcea rosea) 등 주로 아욱과에 속하는 식물에서 감염을 확인하고 보고하였다(Costa 와 Duffus, 1957; Henriques와 Henriques, 1986; Kitajima 등, 1962; Majorana와 Silberschmidt, 1967; Marco, 1975; Menzel 등, 2010; Parrella 등, 2015; Valouzi 등, 2017). 아욱과는 전 세계적으로 200여속 2,300종이 분포하며, 국내에는 어저귀속(Abutilon), 접시꽃속(Althaea), 아욱속(Malva), 무궁화속(Hibiscus), 목화속(Gossypium) 등이 분포하고 있다.
아욱(M. verticillata, curled mallow)은 아욱속에 속하는 주로 온대•아열대 지방에 분포하는 한두해살이풀이며, 우리나라를 포함한 아시아 및 유럽 남부에서 채소 및 한방 재료로 재배되고 있다. 아욱에서 동정된 바이러스는 오이모자이크바이러스(Cucumber mosaic virus, CMV)와 수박모자이크바이러스(Watermelon mosaic virus, WMV) 2종이며, 2015년 중국, 2018년 국내에서 각각 보고하였다(Kim 등, 2019; Peng 등, 2015). 본 연구는 2017년 9월 충남 예산에서 엽맥투명 등의 바이러스 증상을 보이는 아욱을 채집하여 전자현미경 검경, reverse transcription polymerase chain reaction (RT-PCR), 염기서열 확인을 통해 국내 미보고 MVCV를 동정하였고, 바이러스를 순수 분리하여 생물학적, 분자생물학적 분석을 통해 그 특성을 보고하고자 한다.

바이러스의 동정

충남지역 바이러스 조사과정 중 아욱의 잎에 뚜렷한 엽맥투명 및 황화 등 바이러스 증상이 관찰되어 아욱 5주를 채집하였다(Fig. 1). 바이러스 입자를 관찰하기 위하여 병징이 있는 잎의 조직을 딥(Quick DIP) 방법으로 2.5% phosphotungstic acid로 염색한 다음, 투과전자현미경 (LEO 912AB, Carl Zeiss, Jena, Germany)으로 관찰하였다. 그 결과 아욱 5주 모두에서 825-930 nm 크기의 사상형 입자가 확인되었다(Fig. 2A). 국내•외 아욱 및 아욱과에 보고된 바이러스 중 감염이 의심되는 CMV, MVCV, 순무모자이크바이러스 (Turnip mosaic virus, TuMV), WMV 4종에 대한 RT-PCR을 실시하기 위해서 게놈 추출 키트(BCS Plant RNA Prep Kit, Biocube System Co., Gwacheon, Korea)를 이용하여 시료 5점 각각에 서 total RNA를 추출하였다. 4종의 바이러스에 대한 진단용 프라이머 및 RT-PCR premix (SuPrimeScript RT-PCR Premix [2x], GeNet Bio, Daejeon, Korea)를 이용하여 RT-PCR을 실시하였다(Table 1). PCR 산물을 1% agarose gel에서 전기영동을 한 결과, MVCV 특이 프라이머로 증폭된 total RNA 5점에서 약 600 bp의 분자 크기를 갖는 밴드가 확인되었다(Fig. 2B). 여기서 얻어진 PCR 산물 3개를 선발하여 정제 후 direct sequencing으로 Genotech (Daejeon, Korea)에 의뢰하여 염기서열을 결정하였다. 미국 국립생물정보센터(National Center for Biotechnology Information, NCBI)의 BLAST 검색 결과, 기존 GenBank에 등록된 MVCV 분리주들과 84-99%의 높은 염기서열 상동성을 보였고, 양파황화위축바이러스(Onion yellow dwarf virus, OYDV) 및 완두종자전염모자이크바이러스(Pea seed-borne mosaic virus, PSbMV) 분리주들과는 72-74%의 상대적으로 낮은 상동성을 보였다. MVCV를 분리하기 위해서 아욱 5점의 이병엽을 0.01 M 인산완충액(sodium phosphate buffer, pH 7.0)으로 각각 마쇄 후, 아욱, 붉은명아주(Chenopodium amaranticolor), 흰명아주(C. quinoa)에 즙액 접종하여 병징을 관찰하였다. 약 2주 후 아욱의 상엽에서 엽맥투명 및 황화 증상이 관찰되었고, 붉은명아주, 흰명아주의 경우 접종 엽에서만 국부 괴사반점이 관찰되었다. 이러한 병징이 MVCV에 의한 것인지 확인하기 위해 RT-PCR을 실시하였고, 그 결과 모두 양성으로 확인되었다. 붉은명아주와 흰명아주을 이용하여 단일국부병반을 3회 분리 후 전신감염 기주인 아욱에 증식하여 MVCV-Cm1-5으로 명명하였고, 국립농업과학원 미생물은행(KACC)에 바이러스 생물자원으로 기탁하였다(CV180611-1-5). 충남지역의 엽맥투명 및 황화 등의 바이러스 의심 증상을 보인 아욱에서 바이러스 입자관찰, 염기서열 확인, 건전한 아욱에서의 병징 재현을 통해 MVCV를 동정하였고, 국내 아욱에서 MVCV 감염을 처음 확인하였다.

기주식물 반응 및 병징 특성

국내 MVCV 분리주에 대한 기주범위 및 병징 특성을 조사하기 위해 Cm1, Cm3, Cm5를 7과 23종의 지표식물에 접종 후 약 3주간 병징을 관찰하였다. 고추(Capsicum annuum), Nicotiana benthamiana, 땅꽈리(Physalis angulata), 토마토(Solanum lycopersicum) 등 가지과 9종, 호박(Cucurbita moschata), 멜론(Cucumis melon) 등 박과 4종, 번행초(Tetragonia expansa), 천일홍(Gomphrena globosa), 갓(Brassica Juncea) 등 16종 지표식물에 대한 육안 및 RT-PCR 검정 결과 모두 음성으로 나타났다. 아욱과 5종에 대한 생물검정 결과, 아욱에 전신 감염하였으나, 오크라(Abelmoschus esculentus), 히비스커스(Hibiscus), 부용화(H. mutabilis)에서는 접종엽, 상엽 모두 감염하지 않는 특성을 보였다(Table 2). 접시꽃에서는 Cm3만 상엽에서 퇴록반점(chlorotic local) 및 모자이크 증상을 나타냈었고, Cm1, Cm5는 감염하지 못하여 분리주간 차이를 보였다(Fig. 3). 명아주과 2종에서는 MVCV 세 분리주 모두 접종엽에 국부 반점을 형성하였다. 국내 아욱 분리주는 7과 23종 중 3종에 국부 또는 전신 감염하여 좁은 기주범위를 보였으며, 접시꽃에서 분리주 간 차이를 보였다. 독일지역의 접시꽃에서 분리된 MVCV (PV-0963, GQ856544)는 흰명아주(C. quinoa)의 접종 엽에 국부 반점을 형성하여 국내 분리주와 유사한 특징을 보였다(Menzel 등, 2010). 그러나 스페인(MVCV-01-06) 및 이탈리아(Ds-Ba-01, Napoli)의 MVCV 분리주들은 접시꽃과 흰명아주에 감염이 되지 않는 것으로 보고되어 국내 분리주와 다른 기주범위 특성을 보였다(Lunello 등, 2009; Parrella 등, 2015).

염기서열 및 계통 분석

Cm1, Cm3, Cm5 분리주의 외피단백질(coat protein) 영역의 염기서열을 분석하기 위해 NCBI에 등록된 MVCV 염기서열을 이용하여 프라이머(MVCV-CP-2F, 5´-GCTTGGGGTTATGAYGAYCTTG-3´; MVCV-CP-1R, 5´-GTCCTAGGCTAGCGGGATGCAAGT-3´)를 디자인하여 RT-PCR을 실시하였고, PCR 산물은 Expin ComboGP (GeneAll, Seoul, Korea)를 이용하여 정제 후, 이를 pGEM-T Easy vector (Promega, Madison, WI, USA)로 클로닝 하였다. 분리주 당 콜로니 3개를 선발 후 Plasmid DNA를 Genotech에 의뢰하여 염기서열을 얻었다. MEGA-X 프로그램의 DNA alignments를 통해 약 1,286-1,299개의 최종 염기서열을 결정하고, NCBI에 등록하였다(GenBank accession nos. MH290174, MH290175, MH290176). 기존에 NCBI GenBank에 등록된 MVCV 19개 분리주를 이용하여 Geneious Primer를 통해 염기서열 분석결과, 국내 세 분리주은 서로 100% 상동성을 보였고, 멕시코의 토마토에서 분리된 MVCV 분리주(FJ561293)와 99%로 가장 높은 상동성을 보였다. 그리고 독일의 접시꽃에서 분리된 MVCV 분리주(GQ856544)와 84%로 가장 낮은 상동성을 보였다. 그 중 전체 외피단백질 영역이 등록된 MVCV 16 분리주와 Potyvirus속의 PSbMV 외피단백질 염기서열을 outgroup으로 사용하여 국내 아욱 3 분리주의 외피단백질 유전자와 아미노산 수준에서 계통학적 유연관계를 분석을 하였다. Group은 스페인의 Malva sp. (EU884405) 분리주를 제외하고 크게 두 그룹으로 나뉘었으며, group I은 네덜란드의 애기부용(FJ53908), 중국의 접시꽃(KX426562-3, MN116683, KX619649, KX462994), 이탈리아의 당아욱(FM212972, LN651191), 이란 및 스페인의 Malva sp. (KY653927, EU884406-8) 분리주를 포함하였다. Group II는 한국의 아욱(MH290174-6), 스페인의 Malva sp. (EU884409-10), 멕시코의 토마토(FJ561293) 분리주를 포함하며, 이 그룹은 아미노산 수준에서 95-99% 유사한 상동성을 보였다(Fig. 4). 염기서열 및 계통 분석 결과 스페인의 Malva sp. 분리주들을 제외하고는 그룹 안에서 지역 및 기주에 따라 가까운 그룹화가 되는 것을 확인할 수 있었다. 그러나 국내 아욱 세 분리주는 토마토를 감염하지 않지만, 멕시코의 토마토 분리주와 가장 유연관계가 가까웠으며, 아미노산 서열은 99%의 높은 상동성을 나타냈다. 또한 접시꽃과 흰명아주를 감염하지 않는 스페인의 Malva sp. 분리주(Eu884409-10)와 계통학적으로 높은 유연관계를 보여, 이러한 계통학적 결과는 병원성 및 기주 범위와는 연관이 없는 것으로 생각된다. 추후 병원성 차이 및 분리주 간의 접시꽃 등 기주 범위의 규명을 위해 전체 염기서열 분석 등의 추가적인 연구가 진행되어야 할 것이다.

요 약

2017년 9월 충남 예산지역의 아욱 잎에서 엽맥퇴록 및 황화 등 바이러스 증상 관찰되었다. 증상이 있는 아욱 5주에서 핵산을 추출하여 아욱엽맥투명바이러스(Malva vein clearing virus, MVCV)를 포함한 4종 바이러스 특이 프라이머를 이용하여 reverse transcription polymerase chain reaction 수행하였다. MVCV 특이 프라이머로 증폭된 total RNA 5점에서 600 bp의 분자크기를 갖는 밴드가 확인되었다. MVCV 확인하기 위하여 여기서 얻어진 PCR 산물 3개를 정제 후 direct sequencing으로 염기서열을 결정하였다. BLAST 검색 결과, 멕시코의 토마토에서 분리된 MVCV 분리주와 99%로 가장 높은 상동성을 보였다. 명아주속 식물을 이용하여 단일국부병반을 3회 분리 후 Cm1-5으로 명명하였다. Cm1, Cm3, Cm5 분리주를 23종의 지표식물에 즙액 접종하였다. Cm3 분리주는 접시꽃에 퇴록반점 및 모자이크 증상을 나타내었다. 국내 아욱 3 분리주를 포함한 6개 다른 국가 및 식물 종에서 분리된 19 MVCV 분리주들에 대한 외피 단백질 유전자의 계통학적 유연관계분석 결과, 지리적 기원 또는 병원성과는 관련되지 않았다. 본 연구는 국내 아욱에서 MVCV의 자연 발생 및 3 분리주의 특성에 대한 첫 보고이다.
